Nitisinone is a hydroxyphenylpyruvate dioxygenase inhibitor used as an adjunct to dietary restrictions for the treatment of hereditary tyrosinemia type 1 (HT-1), which causes intolerance to tyrosine containing foods.

Nitisinone acts as an inhibitor of the enzyme 4-hydroxyphenyl pyruvic acid and therefore can reduce the formation of HGA. Based on this action, it can be expected that treatment of AKU patients with nitisinone may therefore reduce the consequences of the disorder, especially the joint destruction and the ochronosis of for instance the eye which develops with long-standing disease
